This mixture is then added to a PIP3-coated microplate where competitive binding occurs. Detection is achieved using a peroxidase-linked secondary reagent along with a colorimetric substrate. The resulting colorimetric signal inversely correlates with the amount of PIP3 extracted from the samples.PIP3 production by class I PI3-kinases (PI3-K) plays a crucial role in various cell signaling pathways. Traditionally measuring PI3-K activity involved labor-intensive methods using 32P-labeled substrates and thin-layer chromatography.
