The Cell and Tissue lysates (40ug) were resolved by SDS-PAGE, transferred to PVDF membrane and probed with anti-human PGP9.5 antibody (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection system.
Lane 1.: Mouse Brain lysate Lane 2.: U87-MG cell lysate
ICC/IF analysis of PGP9.5 in U87MG cells line, stained with DAPI (Blue) for nucleus staining and monoclonal anti-human PGP9.5 antibody (1:100) with goat anti-mouse IgG-Alexa fluor 488 conjugate (Green).
Paraffin embedded sections of human nerve tissue were incubated with anti-human PGP9.5 (1:100) for 2 hours at room temperature. Antigen retrieval was performed in 0.1M sodium citrate buffer and detected using Diaminobenzidine (DAB).

PGP9. 5/UCH-L1 is a member of a gene family whose products hydrolyze small C-terminal adducts of ubiquitin to generate the ubiquitin monomer. PGP9. 5 is a component of the ubiquitin system, which has a fundamental role in regulating various biological activities. PGP9. 5 gene encodes two opposing enzymatic activities that affect alpha-synuclein degradation and Parkinson's disease susceptibility.
