| 概要 |
The Sphingosine Kinase Activity Assay is a luminescent assay that measures sphingosine kinase activity by detecting the decrease of ATP.Sample Type: purified enzyme cell lysate/homogenateSample Volume: 10 AμL / sample (duplicate points)Assay Incubation Time: 30 minutes to 2 hoursProduct BackgroundThe Sphingosine Kinase Activity Assay is prepared with the addition of sphingosine kinase sphingosine (substrate) in reaction buffer and initiated by the addition of ATP. The reaction is stopped by addition of the ATP detector at the chosen reaction time. The luminescent signal is inversely correlated with the kinase activity. The sphingosine kinase activity assay was tested with Echelon's SPHK1 and SPHK2.Sphingosine kinases (SPHKs) catalyze the phosphorylation of sphingosine to sphingosine-1-phosphate (S1P). SPHKs have been implicated in proliferation survival migration and regulation of Ca2+ homoeostasis development and regulation of the cardiovascular and nervous systems inflammation immunity and cancer growth. There are two currently known SPHK isoforms SPHK1 and SPHK2.
